Diplomová práca (Ing.). - Ústav aplikovanej biológie FBP SPU v Nitre Subject-s diplomové práce Annotation Autorský abstrakt: Trans-anetol aromatická organická zlúčenina nachádzajúca sa v mnohých koreninách, vrátane badiánu (Illicium verum), anízu (Pimpinella anisum) a feniklu (Foeniculum vulgare) sa vyznačuje sa vysokou biologicky aktivitou. Diplomová práca je zameraná na sledovanie účinku trans-anetolu na kvalitatívne parametre spermií moriakov počas in vitro skladovania. Vzorky boli pristrojené ako melanž ejakulátu s 9 roztokmi rozmanitej koncentrácie trans-anetolu : 15, 10, 7,5, 5, 3,752, 2,5, 1,8752, 1,25 a 0,9375 mmol.l-1. Prostredníctvom CASA systému sa analyzovali jednotlivé parametre pohyblivosti v 7 kultivačných časoch, v 0, 1, 2, 3 ,4, 5, 6 hodín pri skladovacej teplote 5°C. Koncentrácie trans-anetolu 15 a 10 mmol.l-1, vykazovali počas celej doby kultivácie signifikantné negatívne účinky. Inhibičný účinok bol taktiež zaznamenaný v prípade koncentrácií 7,5 a 5 mmol.l-1. Preukazne stimulačný účinok na jednotlivé sledované parametre pohyblivosti boli detegované u koncentrácií 1,8752 a 1,25 mmol.l-1. V rámci sledovania životaschopnosti spermií, prostredníctvom MTT-testu v časových intervaloch 2, 4 a 6 hodín, signifikantne pozitívny účinok bol detegovaný vo vzorke 3,752 a 2,5 mmol.l-1 len po 2 hodinách. Resazurínové farbenie preukázalo stimulačný účinok trans-anetolu vo vzorke 2,5 mmol.l-1 po 2 aj 4 hodinách. Štatisticky negatívny účinok vzorky 15 a 10 mmol.l-1 sa preukázal aj pri sledovaní membránovej integrity. Naopak výrazný ochranný účinok (p≤0,0001), pred membránovým poškodením, bol zaznamenaný pri koncentráciách 2,5 – 0,9375 mmol.l-1 vo všetkých sledovaných časoch. Záverom môžeme povedať, že napriek tomu, že trans-anetol nemá výrazný stimulačný účinok na pohyblivosť a životaschopnosť spermií, môžeme ho odporúčať ako vhodnú protektívnu látku na vylepšenie existujúcich semenných extenderov. Author’s abstract: Trans-anethole aromatic organic compound contained in many spices, including star anise (Illicium verum), anise (Pimpinella anisum) and fennel (Foeniculum vulgare) is characterized by high biological activity. The diploma thesis is focused on monitoring the effect of trans-anethole on the qualitative parameters of turkey sperm during in vitro storage. The samples were prepared as a melange of ejaculate with 9 solutions of various concentrations of trans-anethole: 15, 10, 7,5, 5, 3,752, 2,5, 1,8752, 1,25 and 0,9375 mmol.l-1. CASA system, individual mobility parameters were analyzed at 7 cultivation times, at 0, 1, 2, 3, 4, 5, 6 hours at a storage temperature of 5°C. Concentrations of trans-anethole 15 and 1 mmol.l-1, significant negative effects throughout the cultivation period. The inhibitory effect was also recorded in the case of concentrations of 7,5 and 5 mmol.l-1. A demonstrable stimulatory effect on individual motility parameters was detected at concentrations of 1,8752 and 1,25 mmol.l-1. As part of the monitoring of sperm viability, through the MTT-test at time intervals of 2, 4 and 6 hours, a significantly positive effect was detected in the sample of 3,752 and 2,5 mmol.l-1 after only 2 hours. Resazurin staining demonstrated the stimulating effect of trans-anethole in the sample of 2,5 mmol.l-1 after 2 and 4 hours. The statistically effective effect of the 15 and 10 mmol.l-1 sample was also demonstrated when monitoring the integrity of the membrane. On the contrary, a significant protective effect (≤0,0001) against membrane failure was recorded at concentrations of 2,5 – 0,9375 mmol.l-1 at all observed times. In conclusion, we can say that despite the fact that trans-anethole does not have a significant stimulating effect on sperm motility and viability, we can recommend it as an important protective substance to improve existing sperm extenders.
