Bakalárska práca (Bc.). - Ústav aplikovanej biológie FBP SPU v Nitre Predmet bakalárske práce Anotácia Autorský abstrakt: Bakalárska práca sa venuje problematike energetického zabezpečenia a stability spermií žrebcov v podmienkach in vitro so zameraním na účinky experimentálnych koncentrácií dextrózy. Hlavným cieľom výskumu bolo objasniť vplyv rôznych dávok tohto monosacharidu (v rozsahu od 3,75 mg.ml⁻¹ do 120 mg.ml⁻¹) na kinetické vlastnosti a metabolickú odozvu samčích pohlavných buniek. Kvalita ejakulátu bola monitorovaná v pravidelných časových intervaloch (0, 1, 2 a 3 hodiny) pri teplote 5 °C, čím sa simulovali podmienky krátkodobého skladovania inseminačných dávok. Na objektívne zhodnotenie kvality boli využité dve kľúčové laboratórne metodiky. Kinetické parametre, vrátane celkovej a progresívnej motility, boli analyzované pomocou počítačového systému CASA. Zároveň bola sledovaná mitochondriálna aktivita buniek prostredníctvom kolorimetrického MTT testu, ktorý reflektuje enzymatickú činnosť mitochondrií a celkovú životaschopnosť spermií. Výsledky experimentu ukázali, že bezprostredne po pridaní dextrózy (čas 0 h) nedošlo k štatisticky významným zmenám v sledovaných parametroch a mitochondriálna aktivita všetkých skupín sa udržiavala na vysokej úrovni (90 – 100 %) v porovnaní s kontrolou. Avšak s narastajúcim časom inkubácie bol zaznamenaný postupný pokles hodnôt motility a kinetiky, pričom najvýraznejšia degradácia bola pozorovaná pri najvyšších koncentráciách dextrózy (60 a 120 mg.ml⁻¹). Tento jav pravdepodobne súvisí s nárastom osmotického tlaku v mimobunkovom prostredí, čo vedie k narušeniu membránovej integrity a energetickému vyčerpaniu buniek. Závery práce zdôrazňujú, že hoci je dextróza nevyhnutným substrátom pre tvorbu ATP a podporu motility, jej nadmerná koncentrácia pôsobí na spermie žrebcov inhibične. Získané údaje poskytujú dôležité teoretické aj praktické východiská pre optimalizáciu zloženia inseminačných médií. Author’s abstract: This bachelor thesis addresses the issues of energy supply and stability of stallion spermatozoa under in vitro conditions, focusing on the effects of experimental concentrations of dextrose. The main objective of the research was to clarify the influence of various doses of this monosaccharide (ranging from 3.75 mg.ml⁻¹ to 120 mg.ml⁻¹) on the kinetic properties and metabolic response of male germ cells. The quality of the ejaculate was monitored at regular time intervals (0, 1, 2, and 3 hours) at a temperature of 5 °C, simulating the conditions of short-term storage of insemination doses. Two key laboratory methodologies were utilized for an objective quality assessment. Kinetic parameters, including total and progressive motility, were analyzed using the Computer-Assisted Semen Analysis (CASA) system. Simultaneously, the mitochondrial activity of the cells was monitored via the colorimetric MTT assay, which reflects the enzymatic activity of the mitochondria and the overall viability of the spermatozoa. The experimental results showed that immediately after the addition of dextrose (time 0 h), there were no statistically significant changes in the monitored parameters, and the mitochondrial activity of all groups remained at a high level (90 – 100%) compared to the control. However, with increasing incubation time, a gradual decline in motility and kinetic values was recorded, with the most significant degradation observed at the highest concentrations of dextrose (60 and 120 mg.ml⁻¹). This phenomenon is likely related to an increase in osmotic pressure in the extracellular environment, leading to the disruption of membrane integrity and energy exhaustion of the cells. The conclusions of the work emphasize that although dextrose is an essential substrate for ATP production and motility support, its excessive concentration has an inhibitory effect on stallion spermatozoa.
